Yes, you shouldn't have a problem with your video since Little Reader supports .mp4 files (for your reference, typical formats supported are .avi, .mpg, .mpeg, .mp4 and .wmv)
Can you try installing the latest version of Little Reader and see if that helps? Simply go to Step 1 of this page to download your software installer:

To create a Diagnostic Report:
- From your START Menu, go to 'All Programs'
- Open the 'BrillKids' folder
- Open the 'Little Reader' folder
- Find and run the 'Diagnostic' tool
- A black window will display collecting your system information. Please be patient during this process. - Once the Diagnostic tool has finished, it will open up a window containing a file named "Little Reader_[DATE].zip"
- Attach this file to your reply (simply click on the 'Choose File' button at the bottom of your post composition page and locate this file)
